Identification of N-acylethanolamines in Dictyostelium discoideum and confirmation of their hydrolysis by fatty acid amide hydrolase [Research Articles]

In this study, we provide a further characterization of FAAH and identify NAEs in D. discoideum for the first time. We also demonstrate the ability to modulate their levels in vivo through the use of a semispecific FAAH inhibitor and confirm that these NAEs are FAAH substrates through in vitro studies. We believe the demonstration of the in vivo modulation of NAE levels suggests that D. discoideum could be a good simple model organism in which to study NAE-mediated signaling. Authors: Nathoo S, Litzenberger JK, Bay D, Turner RJ, Prenner EJ Abstract Understanding lipid-protein interactions to enhance our knowledge of membrane architecture is a critical step in the development of novel therapeutic measures to respond to the drastic rise of drug resistant microorganisms. Escherichia coli contains a small archetypal inner membrane multidrug resistance protein, EmrE, that must multimerize to be functional but this multimerization is difficult to demonstrate in vivo. We studied three major E. coli lipids (phosphatidylethanolamine, phosphatidylglycerol and cardiolipin) that varied in head group structure, acyl chain length and saturation. Sphingomyelin synthase (SMS) produces sphingomyelin while consuming ceramide (a negative regulator of cell proliferation) and forming diacylglycerol (DAG) (a mitogenic factor). Therefore, enhanced SMS activity could favor cell proliferation. To examine if dysregulated SMS contributes to leukemogenesis, we measured SMS activity in several leukemic cell lines and found that it is highly elevated in K562 chronic myelogenous leukemia (CML) cells. The increased SMS in K562 cells was caused by the presence of Bcr-abl, a hallmark of CML; stable expression of Bcr-abl elevated SMS activity in HL-60 cells while inhibition of the tyrosine kinase activity of Bcr-abl with Imatinib mesylate decreased SMS activity in K562 cells. Prediction of the goat milk fatty acids by near infrared reflectance spectroscopy

AbstractNear infrared reflectance spectroscopy (NIRS) was used to predict the goat milk fatty acid (FA) profile. The ability of cow milk broad‐based calibration equations to predict the goat milk FA profile was assessed. Three hundred twenty‐eight samples in the calibration set and 108 in the validation set were analysed. We showed that the bias and unexplained error were significant for most of the FA despite an adequate standardized Mahalanobis distance (index to establish the boundaries of a population of samples) which allowed us to test the ability of bovine models to predict goat milk FA profiles.
