Deep eutectic solvents : Synthesis, application and focus on lipase‐catalyzed reactions

AbstractIn recent years, a novel medium with similar properties to Ionic liquids but with additional advantages regarding cost, environmental impact and synthesis has been created: Deep Eutectic Solvents (DESs). These solvents result from the association between an organic salt (ammonium or phosphonium) with a hydrogen‐bond donor such as alcohols, acids or amides. To date, the availability of green, inexpensive and easy to handle solvents is almost nonexistent. Therefore, DESs currently arouse growing interest in many research fields. This review deals with the major applications of this new family of solvents with a particular focus on lipase‐catalyzed reactions such as hydrolysis, aminolysis or alcoholysis. Helfrich's pioneering study of smectic A liquid crystals in 1973, continuum elastic models (CEMs) have been employed as tools to understand the energetics of protein-induced lipid bilayer deformations. Among the assumptions underlying this use is that all relevant protein-lipid interactions can be included in the continuum representation of the protein-bilayer interactions through the physical parameters determined for protein-free bilayers and the choice of boundary conditions at the protein/bilayer interface. Comparison of the pharmacological profiles of murine antisense oligonucleotides targeting apolipoprotein B and microsomal triglyceride transfer protein [Research Articles]

Therapeutic agents that suppress apolipoprotein B (apoB) and microsomal triglyceride transfer protein (MTP) levels/activity are being developed in the clinic to benefit patients who are unable to reach target LDL-C levels with maximally tolerated lipid-lowering drugs. To compare and contrast the metabolic consequences of reducing these targets, murine-specific apoB or MTP antisense oligonucleotides (ASOs) were administered to chow-fed and high fat-fed C57BL/6 or to chow-fed and Western diet-fed LDLr–/– mice for periods ranging from 2 to 12 weeks, and detailed analyses of various factors affecting fatty acid metabolism were performed. Enzymatic hydrolysis of palm stearin to produce diacylglycerol with a highly thermostable lipase

AbstractThis paper firstly presents the production of diacylglycerol (DAG) by hydrolysis of high‐melting oil by a highly thermostable lipase (T1 lipase). T1 lipase was expressed in recombinant Escherichia coli, and used to produce diacylglycerol by catalyzing the hydrolysis of palm stearin. The optimal conditions were obtained by a water content of 30% (w/w, with respect to oil mass), T1 lipase at a concentration of 7.5 U/g (U/w, with respect to oil mass) and at 60oC. Scale‐up reaction was attempted based on the optimized conditions and 28.1% of DAG was achieved. Molecular distillation was employed for purification of DAG from the reaction mixture and the final DAG product yielded 42.5% of DAG (27.8% for 1,3‐DAG and 14.7 for 1,2‐DAG) and 57.5% of triacylglycerol (TAG).
